Nucleic Acid Electrophoresis

DNA electrophoresis is used to separate DNA fragments by size. The types of gels most commonly used for DNA electrophoresis are agarose (for relatively large DNA molecules) and polyacrylamide (for high resolution of short DNA fragments).  The former is also sometimes referred to as submarine electrophoresis because the gels are submerged in buffer. The buffers used for the separation are Tris-Borate-EDTA or Tris-Acetate-EDTA and the DNA migrates relative to its size: smaller fragments moving faster through the gel matrix and larger fragments moving slower. Following the separation, the gels can be stained or transferred (Southern blotting). DNA separations are usually detected by dyes that bind to the molecules and are typically viewed under UV illumination. Nucleic acids can be transferred from the gel matrix to membrane sheets by capillary or electrical transfer for probing.
DNA fragments in the range of 100 bp to 20 kb are typically separated in agarose gels using submarine style electrophoresis units. Hoefer offers 8sizes of submarine units for quick results or high throughput.
Smaller fragments, <100 bp, are typically separated on polyacrylamide gels in a vertical format. We recommend the Hoefer MiniVE or the Mighty Small II units for small fragments up to 1 kb.

Mini Submarine Electrophoresis Units
Hoefer offers a choice of three Mini Submarine units from the SUB6 (6 x 7.5 cm), the SUB10 (10 x 11.5 cm) to the HE33 Mini Submarine Unit (7 x 10 cm) for nucleic acid analysis in the standard size gel range. The SUB6 Mini and SUB15 Mini-Plus Submarine Units are Ideal for routine preparatory and analytical studies of nucleic acids.  The SUB10 provides the most commonly used gel tray size and comb options. The HE33 Mini provides you with the to perform rapid separation of DNA fragments
  • The SUB6 comes complete with Buffer chamber with safety lid and power leads,  UVT running tray (cast and turn; casting) two 1 mm thick  x 16 well combs, colored Loading strips, buffer recirculation ports.
  • The SUB10 comes complete with Buffer chamber with safety lid and power leads,  UVT running tray with silicone gaskets,  two 1 mm thick x 16 well combs, colored Loading strips, buffer recirculation ports.
  • The HE33 comes complete with Buffer chamber assembly, safety cover with power leads, one 7 x 10 cm running tray, casting tray 7 x 10 cm, four foam gaskets, one 1.5 mm thick x 8 well comb,  bubble level.
Standard Submarine Electrophoresis Unit
Hoefer offers a comprehensive line of Standard Submarine units; the SUB13, the SUB15 and the HE99X Max Submarine Unit for nucleic acid analysis in the standard size gel range. The SUB13 Midi Submarine Unit is Ideal for analytical and preparative studies of nucleic acids.  The SUB15 provides the most commonly used gel tray size and comb options.  The HE 99X Max provides you with the flexibility of choosing from a variety of gel lengths and comb sizes so you can design the gel that gives maximum resolution of DNA fragments.

Maxi and Maxi-Plus Submarine Units
Hoefer offers a choice of four large Submarine units: the SUB20 (20 x20 cm) Maxi-Standard  unit, the SUB20C Maxi-Cooled unit, the SUB25 (25 x 30 cm) Maxi-Plus and SUB25C Maxi-Plus Cooled Submarine Unit for nucleic acid analysis.  The SUB20 Maxi-Standard and the SUB20C Maxi-Cooled Submarine Unit are Ideal for high resolution analytical and preparative studies of nucleic acids.  The SUB25 and SUB25C are ideal for high throughput screening of nucleic acids, particularly PCR screening and in genotyping studies.
  • The SUB20 and SUB20C come complete with Buffer chamber, safety lid and power leads,  UVT running tray with casting gates (20 x 20 cm), two 1mm thick x 16 well combs, colored Loading strips, buffer recirculation ports.
  • The SUB25and SUB25C comes complete with Buffer chamber assembly, safety cover with power leads, one 25 x 30 cm running tray with casting gates 15 x 20 cm, Six 1.0 mm thick x 26 well combs.
